4.      Procedure

4.1     DNA-Extraction

Feather samples:

(1)     Get 4-6 feather samples, cut the roots into a centrifuge tube, shake the centrifuge tube to make the feather roots reach the bottom of the tube.

(2)     Add 50ul "Nucleic Acid Release Agent" to the above centrifuge tube.Vortex for 30s，Add 100ul DEPC H2O to the tube. Gently flick the reagent to the bottom of the tube.

4.2      Reagent preparation

Take out the “Lyophilized PCR Reaction Tubes” and add 20ul "Reconstitution Solution", add 5ul sample to be tested. Take out the “PCR Tube Cap” and fasten the cap, Vortex for 10s, instantaneous centrifugation.

4.3      qPCR amplification

Set up the qPCR amplification program:

Fluorescence channel selection: FAM and VIC
