Testing method

 

1.  Detection operation:

Feather samples: ( 1 ) Take 4-6 feather samples, cut the roots into a centrifuge tube, shake the centrifuge tube so that the feather roots reach the bottom of the tube;      ( 2 ) Add 50 ul "nucleic acid release agent" to the above centrifuge tube, vortex for 30 s , and gently shake the liquid to the bottom of the tube, it can be used as samples to be tested. (Samples to be tested should not be stored at 4 °C for more than 1 day)

2.   Reagent preparation:

Take out the lyophilized reagent, add 20 ul "reconstitution solution" +5 ul sample to be tested to the well, cap tightly for PCR Tube cap, vortex to mix, momentary centrifugation. Put the PCR tube into the PCR machine and set up the following programs:

4.   Interpretation of results:

Female : FAM Channel test result is positive, VIC channel test result is positive;

Male : FAM Channel test result is negative, VIC channel test result is positive;

Exception : If the VIC  channel test result is negative, the nucleic acid wasn’t extracted from the sample, and the test should be repeated.
